Analisis Kualitas Air Galon pada Depot Air Minum di Kota Palopo dengan Menggunakan Metode MPN (Most Probable Number)

Air minum aman bagi kesehatan apabila memenuhi persyaratan secara fisika, mikrobiologi, kimia, dan radioaktif. Kualitas air minum di Indonesia harus memenuhi persyaratan yang tertuang di dalam Peraturan Menteri Kesehatan RI No.492/MENKES/PER/IV/2010 dimana setiap komponen yang diperkenankan berada di dalamnya harus sesuai. Parameter wajib penentuan kualitas air minum secara mikrobiologi adalah total bakteri Coliform dan Escherichia coli dilakukan dengan Most Probable Number Test. Jika di dalam 100 ml sampel air didapatkan sel bakteri Coliform, memungkinkan terjadinya diare dan gangguan pencernaan lain. Sampel pada penelitian ini adalah air galon yang diambil dari beberapa depot air minum di Kota Palopo. Sebanyak 1 mL sampel diambil kemudian diinokulasikan kedalam botol yang berisi NaCl fisiologis sebanyak 9,9 mL (seri 10-1). Selanjutnya dibuat seri 10-2 dengan mengambil sampel air yang sebanyak 1 mL kemudian menginokulasikannya kedalam botol yang berisi NaCl fisiologis sebanyak 9,9 mL dan menginokulasikannya pada tiap tabung reaksi 10-1, kemudian dibuat seri 10-3 dengan mengambil sampel sebanyak 0,1 mL lalu menginokulasikannya kedalam tiap-tiap tabung reaksi 10-2 dan pada tiap-tiap tabung reaksi 10-3. Selanjutnya dilakukan pengamatan setiap 12 jam sekali dalam waktu 3 hari lalu mencatat perubahan yang terjadi. Hasil penelitian menunjukkan bahwa ketiga sampel dari tiap seri tabung dengan volume 10 mL positif mengandung bakteri Coliform. Setelah hasilnya dicocokkan dengan tabel MPN seri 9 tabung berdasarkan Bacterial Analytical Manual, didapatkan hasil bahwa jumlah bakteri Coliform pada air galon per 100 ml adalah >1100. Melihat kondisi air galon tersebut dengan nilai Coliform lebih dari 10 per 100 ml, maka air tersebut sudah tidak layak dikonsumsi.
